How many? Dozens,hundreads, thousands? Once or regularly? Are the email addresses validated? Maybe look at the import scripts? Another potential solution is the API.

Thousands. What if they already opted-in? Is there a way to just make them members?

You can do it with the API or a script like the import scripts.
