I’m trying to preserve the oneboxing of the homepage of a specific domain, but prevent the oneboxing of any thing more that that. i.e.
mydomainofinterest.com makes a onebox
mydomainofinterest.com/anything does not
Is this achievable? I can only seem to block an entire domain - which would be fine if I could get the homepage of that domain to still make a nice onebox.

No. It’s not achievable. All subdomains and paths under the blocked domain are blocked too.

The use-case is link based video-conferencing.
