We have a user that we think complies with trust level 2, before we just go ahead and manually give a tl2 to, is there a SQL command or option to force discourse to reassess his TL first?
I have tried changing it down to 0 then back to 1 to force a reassess and also a lock and unlock

I’m not sure what you mean. Users move between Trust levels 0 - 3 automatically (TL4 is manual promotion) when they reach certain requirements, so you don’t have to manually promote users. You can change the requirements in your site settings:
example.discourse.com/admin/site_settings/category/trust?filter=trust%20levels
If you would like the user in question to stay at TL2 you can, as you pointed out, lock their trust level at 2.
