Can you enter the site via Safe Mode?
I think you may need to elaborate. 
Have you updated between ‘working’ and ‘not working’?
Can you do a rebuild from the command line and see if that resolves your issues?
My guess is that you did an upgrade from the web interface and need to do a command line rebuild.
